The Disadvantages of Water Bath Thawing

The most common method for rapidly thawing frozen cell samples is to partially submerge cryovials in a 37 ℃ water bath for a few minutes and then observe the sample to determine if thawing is complete. However, this method has some significant drawbacks.

  • High risk of contamination: Biological samples in the cryovial have a very high chance of becoming contaminated.
  • Not a sterile process: A water bath cannot be used as part of a sterile process.
  • Subjective and inconsistent: Different operators may have significant variations in determining the thawing time, final temperature, and endpoint, as the process relies on experience.
  • Regulatory limitations: The use of water baths is restricted in GMP or clinical settings.
  • Non-standardized: The process is not standardized or intelligent, making it impossible to integrate into automated cell culture equipment.
